Evaluation of S100 A4& A6 protein expression in B-cell chronic lymphocytic leukemia

B-cell chronic lymphocytic leukemia (B-CLL) is the most common leukemia in adults. Its clinical course is extremely heterogeneous, and in some cases it shows an aggressive course. As of now, no potential markers have been identified for the early prognosis of B-CLL. S100A4 and S100A6 are members of S100 family of calcium binding proteins that is emerging as a potentially important group of prognostic markers in cancer development and progression. Although there is considerable evidence that S100A4 and S100A6 genes play important roles in metastasis development and increased the aggressiveness of solid tumors, the expression of S100 A4/A6 proteins in B-cell malignancies is not clearly investigated to date. This study aimed to investigate the expression of S100 A4/A6 in B- cell CLL as a possible marker of stemness, which can be used for the characterization of CLL types and prognosis. The investigation was carried out using immunohistochemistry followed by double labeling immunofluorescence analysis of B- lymphocyte lymphomas and bone marrow samples. Interestingly, the immunohistochemistry results showed a small proportion of B cell lymphomas were S100A4 positive, including 3 CLL samples, and one case of MZL, MCL, and DLBCL, but not in FL or Hodgkin’s lymphomas. Based on immunofluorescence staining S100A4 signals was detected only in 4 cases including 3 CLL cases and case of MZL, but not in MCL or DLBCL. S100A6 protein found to not express in the neoplastic B lymphocytes in the analyzed B-cell lymphomas. Because S100A4 expression was detected in CD34, CD133, CD10 positive, this study corroborates the expression of S100A4 in bone marrow HSCs. Those give indication to the potential function of S100A4 in the maintenance of stemness features of B cell lymphomas. And enhance the possibility of using S100A4 expression as marker in the prognosis of CLL, through further analysis of more samples and
