The antibacterial activity using the agar well diffusion method showed that the novel
AU-23 compound possesses remarkable antibacterial activity against four reference
ATCC strains (Pseudomonas aeruginosa ATCC 9027, methicillin-resistant
Staphylococcus aureus (MRSA) ATCC 33591, methicillin-sensitive Staphylococcus
aureus(MSSA) ATCC 25923, and methicillin-resistant Staphylococcus aureus (MRSA)
ATCC 43300) that commonly cause wound infection, with an average inhibition zone
ranging between 12mm and 18mm. The MIC and MBC of the AU-23 compound against
the ATCC strains ranged from 15.63 to 62.5 µg/mL. Moreover, the dose-dependent AU-
23 exhibited bactericidal activity against two sensitive ATCC bacterial strains, P.
aeruginosa 9027 and MSSA 25923, during the first 4 and 6 hours. Antibiofilm activity
using the XTT assay was significantly higher against both ATCC strains, MSSA 25923
and P. aeruginosa 9027, when grown in mono-microbial biofilm mode, with inhibition
rates of 64.98% and 59.16%, respectively. However, prenylated flavonoid PF-23
showed a slightly good antibacterial effect against MSSA25923 and P. aeruginosa
9027, with inhibition zones ranging from 10 to 11mm, and MIC/MBC values between
500 and 1000 µg/mL.
