| dc.description | Our results revealed that Murashige and Skoog (MS) basal medium was the best medium for plantlets multiplication compared with those grown on Woody Plant Medium (WPM). Regarding growth regulators, our results demonstrated that MS basal medium supplemented with 1mg/l kinetin (kin) revealed the highest number of shoots per explant produced by single node after 8 weeks of culture. The MS basal medium containing 4mg/l. growth vii regulator Indol-3- butyric acid (IBA), however, was the best in term of rooting of growing shoots. | |
| dc.description.abstract | This study was conducted to find out the best time to collect Bupleurum fruticosum L, and to determine the most suitable medium for shoot multiplication and root stimulation as well as to determine the maximum period could plantlets be saved in the fridge for further use in tissue propagation; in addition, plantlets acclimatization process was experienced through gradual transfer from in vitro to the field. To gain plant-tissue free of contamination, experiments were conducted to find the best method for explants surface sterilization. The results illustrated that best method was by soaking explants in 3% chlorox solution for twenty minutes. For explants collected in different periods those were collected in May exhibited lease contaminated explants. | |
